Anti-oxidative and proteolytic activities and protein profile of laticifer cells of Cryptostegia grandiflora, Plumeria rubra and Euphorbia tirucalli Braz. J. Plant Physiol.

In this study, proteins extracted from laticifer cells of three plants were examined by electrophoresis, mass spectrometry (MALDI-TOF) and characterized in respect of proteolytic, chitinolytic and anti-oxidative activities by means of zymography and colorimetric assays. Acidic proteins with molecular masses between 12.5 and 74.5 kDa predominated in laticifers of P. rubra. This profile was not found in laticifers of C. grandiflora and E. tirucalli. The later was poor in respect of proteins. According to the model frequently used as reference, plant cell growth occurs only when turgor surpasses a threshold. This model was proposed considering a cylindrical cell of constant wall thickness immerged in a water solution, with viscoelastic behaviour, hydraulic conductivity, variable extensibility, and unidirectional elongation. The author, Lockhart (1965), did not consider the effects of apoplasm hydrostatic potential, a subject treated later by Calbo and Pessoa (1994) who argued that this component of cell potential would interfere with cell growth rate. Since the first isolation of ascorbic acid (AsA) in 1928, few papers have been published regarding the biosynthesis of AsA in plants, especially in fruits. It took as long as 1998, before Wheeler, Jones and Smirnoff, based on a study with Arabidopsis leaves, proposed what can be considered the main pathway of biosynthesis of AsA, in which L-galactose (L-GAL) is a key precursor. Coffee plants are highly N-demanding plants. Despite the importance of N nutrition for the development, acclimation and yield of coffee plants, there are few reports concerning N metabolism in this species. In this review, our intention is to summarize the information available in the literature and to point out the influence of environmental conditions on N assimilation, as well as comment and discuss some apparently contradictory results and raise and enlighten queries about N assimilation in coffee plants.

Asymmetrical leaves induced by water deficit show asymmetric photosynthesis in common bean Braz. J. Plant Physiol.

In this study we tested the hypothesis that leaf asymmetric growth induced by water deficit in common bean (Phaseolus vulgaris L.) might be correlated with different net photosynthesis rates between the two opposed leaflets (right and left) considering a bilateral symmetric plane. In order to induce developmental instability, the drought-sensitive common bean genotype "Jalo Precoce" was subjected to periods of suspended irrigation during its vegetative growth. Developmental instability was evaluated by measurements of leaflets asymmetry, and the effects of drought on gas exchanges were taken in both symmetrical leaflets (right and left) in relation to the central leaflet. Avaliaram-se os efeitos do estresse hídrico e da reidratação sobre parâmetros de fluorescência, condutância estomática e peroxidação de lipídios em plantas de Myracrodruon urundeuva Fr. All. (Anacardiaceae). As plantas foram cultivadas em casa de vegetação e a seca foi induzida por suspensão da irrigação durante 14 dias. Sob estresse hídrico, observou-se queda acentuada no conteúdo relativo de água foliar, o qual alcançou valores próximos a 60 %. Essa queda foi acompanhada por grande diminuição na condutância estomática e na taxa aparente de transporte de elétrons (ETR) e extensivos danos às membranas evidenciados por aumentos na peroxidação de lipídios. Autonomy and network modulation of photosynthesis and water relations of Coffea arabica in the field Braz. J. Plant Physiol.

The degree of connection between leaf gas exchange and leaf water potential, and the autonomy of these variables in relation to meteorological conditions were determined in three cultivars of Coffea arabica during clear and cloudy days. High values of vapor pressure deficit, air temperature and photosynthetic photon flux density resulted in low leaf autonomy during a clear day, irrespective the degree of connection among leaf physiological variables. Coffee is one of the most valuable agricultural commodities and ranks second on international trade exchanges. The genus Coffea belongs to the Rubiaceae family which includes other important plants. The genus contains about 100 species but commercial production is based only on two species, Coffea arabica and Coffea canephora that represent about 70 % and 30 % of the total coffee market, respectively. O gênero Kohleria sp pertencente à família Gesneriaceae possui órgão subterrâneo. Este órgão é um caule envolto por folhas modificadas, imbricadas, as quais acumulam reservas e cada uma destas protege uma gema. Seções do rizoma brotam sem necessidade do órgão inteiro. Seções deste rizoma com seis folhas modificadas foram colocadas para brotar em placas de Petri e testadas quanto às temperaturas de 15, 20, 25, 30 e 35 ºC, na luz ou no escuro e ainda em relação aos teores de umidade do substrato, onde se adicionaram os volumes de 1, 12 e 20 mL de água destilada. In this study, the effects of cadmium (Cd) on lipid peroxidation, electrolyte leakage, protein oxidation, ascorbate peroxidase (APX; E.C. 1.11.1.11), catalase (CAT; E.C. 1.11.1.6) and superoxide dismutase (SOD; E.C. 1.15.1.1) activities, and ascorbic acid, non-protein thiol groups and total soluble protein contents in cucumber seedlings (Cucumis sativus L.) were investigated. Seedlings were grown in vitro in an agar-solidified substrate containing four Cd levels as CdCl2 (0, 100, 400, and 1000 µmol L-1) for 10 d. The reactive oxygen species generation, lipid peroxidation and antioxidative enzyme response of water hyacinth and salvinia to Cd were evaluated. Cadmium was absorbed/accumulated mainly in the roots, but significant amounts also translocated to the leaves. No Cd effect on dry weight was detected, although toxicity symptoms were visible. Superoxide and H2O2 concentrations increased, in addition to lipid peroxidation in both species, especially in the leaves of salvinia. In general, antioxidative enzyme activities were reduced in both species following Cd treatment, especially in salvinia. Cadmium-Zinc interactions in a hydroponic system using Ceratophyllum demersum L.: adaptive ecophysiology, biochemistry and molecular toxicology Braz. J. Plant Physiol.

The interaction between an essential micronutrient, Zn and a toxic non-essential element, Cd has been comprehensively reviewed based on our experiments conducted with Ceratophyllum demersum L. in a hydroponic system. Since Cd and Zn belong to the group IIB transition elements and show similarities in chemistry, geochemistry and environmental properties, it would be one of the elemental pairs of choice to investigate metal-metal interactions. Evidence in support of the protective role of Zn against Cd toxicity in Ceratophyllum demersum L. is presented in this review. Based on our experimental results, we conclude that the antioxidant properties of Zn play an important role in counteracting Cd toxicity.

O etileno, conhecido, sobretudo, como o fitormônio da maturação de frutos, está envolvido em vários processos fisiológicos, atuando na regulação da expressão de grande número de genes. O clone parcial de cDNA ER49 foi previamente isolado mediante a técnica do "Differential Display". A obtenção e a análise da seqüência completa confirmaram as homologias significativas com fatores de elongação da síntese de proteínas do tipo Ts. Leaves of Artemisia annua L. are a plentiful source of artemisinin, a drug with proven effectiveness against malaria. The aim of this study was to classify the photosynthetic mechanism of A. annua through studies of the carbon isotope composition (delta 13C) and the leaf anatomy. A. annua presented a delta 13C value of - 31.76 ± 0.07, which characterizes the plants as a typical species of the C3 photosynthethic mechanism, considering that the average delta 13C values for C3 and C4 species are -28 and -14, respectively. Studies involving amplified fragment length polymorphism (cDNA-AFLP) have often used polyacrylamide gels with radiolabeled primers in order to establish best primer combinations, to analyze, and to recover transcript-derived fragments. Use of automatic sequencer to establish best primer combinations is convenient, because it saves time, reduces costs and risks of contamination with radioactive material and acrylamide, and allows objective band-matching and more precise evaluation of transcript-derived fragments intensities. Physiological and anatomical features of Cd-tolerance in Pfaffia glomerata were examined by exposing plantlets to nutrient solutions with increasing Cd concentrations (0, 15, 45, and 90 µmol Cd L-1), and possible Cd-tolerance markers were established. Cd contents were found to be higher in roots than in shoots. According to the bio-concentration factor data, this species is effectively a Cd-hyperaccumulator, as previously attested. Cd induced the appearance of xeromorphic characteristics in leaves (decreased water potential, increased numbers and decreased stomata size) and increased root endodermis thickness.
